Description

Perforin 1 is encoded by the PRF1 gene on chromosome 10q22 and is synthesized as a 555-amino-acid glycoprotein (~67 kDa) containing a C-terminal C2 domain that binds target membranes and an N-terminal MACPF (Membrane Attack Complex/Perforin) domain responsible for pore assembly. The CLIA Kit from Assay Genie pairs a pre-coated microplate with luminescent detection chemistry, so signal is read on a luminescent-compatible plate reader rather than at 450 nm. The kit contains 96 assays and is calibrated against recombinant human perforin, with a sandwich-format capture and detection antibody pair that shows no significant cross-reactivity against related granule proteins such as granzyme A or granzyme B.

Sample handling follows standard collection guidance: whole blood should clot for 2 hours at room temperature or overnight at 2–8 °C before centrifugation, while EDTA or heparin plasma is centrifuged at 1,000 x g for 15 minutes at 2–8 °C within 30 minutes of draw. Supernatants can be aliquoted and stored frozen at -20 °C or -80 °C to limit freeze-thaw cycles, and unopened reagents are kept refrigerated at 2–8 °C in their sealed foil pouch protected from light. The assay is for research use only and is commonly cited in studies of familial hemophagocytic lymphohistiocytosis, pediatric immunodeficiency, Epstein-Barr virus–associated lymphoproliferative disease, graft-versus-host disease after hematopoietic stem cell transplantation, and tumor immune surveillance.
