Human Presenilin-1 ELISA Kit (HUFI07066)

From Assay Genie, this Sandwich ELISA Kit is designed for the precise quantification of Presenilin-1 levels in human samples, including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. The kit uses a colorimetric, double-antibody sandwich detection method with a sensitivity of 0.188 ng/mL and a range of 0.313–20 ng/mL. Presenilin-1 (PSEN1) is a key protein involved in processes related to Alzheimer's disease, including the production of amyloid beta peptides and the regulation of neuronal cell signalling, making accurate measurement of it valuable for research into Alzheimer's disease and other neurodegenerative disorders.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks, and incubating the pre-coated plate at 37°C for 90 minutes to allow antigen binding. A biotin-labelled detection antibody is then added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes at 37°C. TMB substrate is added and incubated in the dark for 10–20 minutes, after which stop solution is added and absorbance is read at 450 nm. Linearity in serum (n=5) ranges from 85–97% at 1:2 to 80–98% at 1:8, in EDTA plasma (n=5) from 86–103% at 1:2 to 84–96% at 1:8, and in heparin plasma (n=5) from 88–105% at 1:2 to 80–95% at 1:8. Recovery averages 93% for serum (range 87–99%), 98% for EDTA plasma (range 90–104%) and 98% for heparin plasma (range 91–104%). The kit should be stored at 2–8°C for 12 months.
