Human precursor of Prostate Specific/proPSA ELISA Kit

Designed for the quantitative measurement of proPSA in human research samples, this Sandwich ELISA Kit from Assay Genie uses a double-antibody configuration and a colorimetric detection approach. ProPSA is a precursor to PSA (Prostate-Specific Antigen) and is studied as a biomarker relevant to prostate cancer development and progression.

The kit has a sensitivity of 0.281 ng/mL and a working range of 0.469-30 ng/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported linearity in serum (n=5) ranges from 85-101% (1:2), 86-100% (1:4) and 80-100% (1:8); EDTA plasma (n=5) shows 86-105% (1:2), 83-97% (1:4) and 80-97% (1:8); and heparin plasma (n=5) shows 86-101% (1:2), 82-100% (1:4) and 83-99% (1:8). Recovery ranges are 85-98% (average 93%) for serum (n=5), 95-103% (average 99%) for EDTA plasma (n=5), and 86-104% (average 93%) for heparin plasma (n=5). Components are stored at 2-8°C, with select items suitable for 12 months at -20°C where indicated.

The assay follows a six-step protocol: equilibrate reagents and set up standards, samples and blanks on the pre-coated plate, then incubate at 37°C for 90 minutes; add biotin-labeled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; add TMB substrate and incubate in the dark for 10-20 minutes; then add stop solution and read absorbance at 450 nm.
