Human PR3 (Proteinase 3) CLIA Kit (HUES01073)

PR3 is encoded by the PRTN3 gene on chromosome 19p13.3 and circulates in both soluble and membrane-bound forms on activated neutrophils. Beyond its classical link to granulomatosis with polyangiitis, dysregulated PR3 expression contributes to microscopic polyangiitis, eosinophilic granulomatosis with polyangiitis, and anti-GBM disease, and circulating PR3 also participates in neutrophil extracellular trap (NET) formation. The Assay Genie CLIA Kit uses a two-site sandwich configuration in which a capture antibody is pre-coated on the microplate; after sample incubation, a biotinylated detection antibody binds, followed by streptavidin-conjugated acridinium ester that emits light upon oxidation. A typical run takes about 2.5 hours at room temperature with sample volumes around 100 µL per well. The 96-well microplate is supplied with lyophilized standards that are reconstituted in assay diluent to generate an eight-point calibration curve, and quality controls at low, mid, and high concentrations are used to verify reproducibility. Researchers can also titrate synovial fluid, bronchoalveolar lavage, or cell culture supernatant, and the assay pairs well with downstream Western blot validation. Reagents remain stable at 2–8 °C until the kit expiration, while reconstituted standards should be aliquoted and frozen at -20 °C or below to limit freeze-thaw degradation.
