PPT1 is a lysosomal thioesterase that cleaves thioester-linked fatty acyl groups from cysteine residues of S-acylated proteins; loss-of-function variants in the PPT1 gene cause CLN1 disease, the infantile form of neuronal ceroid lipofuscinosis (Batten disease), characterized by progressive neurodegeneration, retinal degeneration, and early-onset seizure activity.
