Description

From Assay Genie, this is a Sandwich ELISA designed for the quantitative measurement of human Peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PPARGC1A). The kit shows a sensitivity of 0.188 ng/mL and a working range of 0.313-20 ng/mL (S1). Detection is colorimetric, and the format is a Sandwich ELISA, Double Antibody assay (S1). Acceptable sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples (S1).

Assay performance has been characterised for serum, EDTA plasma and heparin plasma. Reported spike-and-recovery averages are 97% for serum, 98% for EDTA plasma and 92% for heparin plasma, with corresponding ranges of 91-103%, 96-102% and 86-104% (S1). Linearity in serum at 1:2, 1:4 and 1:8 dilutions is 88-97%, 93-105% and 92-103%; EDTA plasma shows 84-101%, 84-100% and 86-99%; heparin plasma shows 88-99%, 82-96% and 83-98% (S1). For storage, the manufacturer recommends 2-8°C with a shelf life of 12 months (S1). Synonyms listed for the target include PPARGC1A, PGC-1-alpha, PPAR-gamma coactivator 1-alpha, PPARGC-1-alpha, Ligand effect modulator 6, LEM6, PGC1, PGC1A, PPARGC1 and PGC-1v (S1).

The protocol involves equilibrating reagents and the TMB substrate to room temperature, loading standards, samples and a zero control onto the pre-coated plate, incubating for 90 minutes at 37°C, adding a biotin-labelled detection antibody for 60 minutes at 37°C, then HRP-Streptavidin for 30 minutes at 37°C, developing with TMB substrate for 10-20 minutes in the dark, stopping the reaction and reading absorbance at 450 nm immediately (S1). The target is associated with research into energy metabolism, mitochondrial biogenesis, metabolic disorders, neurodegenerative diseases and aging (S1).
