Description

This Sandwich ELISA kit from Assay Genie is designed for the precise quantitative measurement of Polycomb group RING finger protein 6 (PCGF6) in human samples. PCGF6 is a key transcriptional repressor involved in chromatin remodeling and gene expression regulation, functioning as a component of a Polycomb group (PcG) PRC1-like complex that maintains the transcriptionally repressive state of many genes, including Hox genes, throughout development. Dysregulation of PCGF6 has been implicated in various diseases, including cancer and developmental disorders, making it a valuable biomarker for studying disease mechanisms and potential therapeutic strategies.

The kit is reactive with human samples and validated for use with serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids. It employs a colorimetric ELISA detection method and detects both natural and recombinant human PCGF6. The assay has a detection range of 78-5000pg/mL and a sensitivity of 20pg/ml, with an intra-assay CV of 5.6% and an inter-assay CV of 9.2%. Linearity has been demonstrated across serum (110-118% at 1:2, 103-113% at 1:4, 110-118% at 1:8, 106-114% at 1:16), EDTA plasma (102-111%, 87-96%, 101-111%, 100-110%), and heparin plasma (91-100%, 103-116%, 96-106%, 92-105%) dilution series. Recovery averages 98% for serum (range 92-104%) and 100% for plasma (range 94-106%).

PCGF6, also known by the synonyms Mel18 and Bmi1-like RING finger, RING finger protein 134, MBLR, and RNF134, is a nuclear protein (UniProt: Q9BYE7) with a molecular weight of 30,249 Da, encoded by the PCGF6 gene located at chromosomal position 10q24.33. It may modulate the levels of histone H3K4Me3 by activating the KDM5D histone demethylase and interacts with BMI1/PCGF4, RING1, RNF2, KDM5D, and CBX4, CBX6, CBX7, and CBX8. The kit is intended for research use only and supports studies within the epigenetics research area, including investigations into gene regulation, chromatin biology, and disease pathology.
