Built around a pre-coated microplate and supplied as a complete assay package, this in vitro research-use ELISA kit from Assay Genie quantitatively measures RNA Polymerase II Subunit I (RPB9), the conserved small subunit of human RNA Polymerase II. POLR2I contributes to transcription start-site selection and elongation fidelity, and quantifying its levels supports studies of transcriptional regulation, chromatin-associated RNA biogenesis, and disease contexts where the subunit has been implicated, including craniofacial development disorders such as Treacher Collins syndrome, in which hypomorphic POLR2I variants have been described.

Each kit is sized for 96 individual determinations on a single plate format and ships with calibrated recombinant standards, biotinylated detection reagents, streptavidin-HRP conjugate, wash buffer, substrate, and stop solution sufficient for the full plate. The product is intended for laboratory research use only and is not validated for clinical diagnostic decision-making. Typical workflows involve preparing clarified samples, running a standard curve, and reading absorbance to interpolate POLR2I concentrations in experimental lysates.

Applications include comparing POLR2I protein abundance between treated and untreated cell populations, profiling transcription-machinery levels across patient cohorts, validating transcriptomic findings at the protein level, and supporting mechanistic studies of how the RPB9 subunit interfaces with the larger Pol II complex during initiation and elongation within an in vitro research framework.
