Background on PNPLA3 and the Assay

Adiponutrin, also called PNPLA3 (patatin-like phospholipase domain-containing protein 3), is a liver-enriched enzyme with patatin-domain phospholipase activity that hydrolyzes triglycerides and retinyl esters on intracellular lipid droplets. The protein is a 481-amino-acid, approximately 53 kDa membrane-bound enzyme, and its circulating concentration is increasingly studied as a biomarker of hepatic steatosis, insulin resistance, and non-alcoholic fatty liver disease progression. The well-characterized I148M (rs738409) polymorphism strongly influences hepatic lipid handling, making quantitative PNPLA3 measurement valuable for translational hepatology research.

Kit Configuration and Workflow

Assay Genie's Human PNPLA3 ELISA Kit ships as a complete pack sufficient for 96 determinations, configured around a pre-coated microplate. Each kit includes recombinant human PNPLA3 calibrators, a biotinylated detection antibody, substrate solution, concentrated wash buffer, assay diluents, and pre-coated microwell strips in individual vials. Standard curves are generated against recombinant calibrators, and absorbance readings are taken on a standard plate reader. Reagents should be stored refrigerated at 4 degrees C; reconstituted standards should be aliquoted for single use and protected from light to preserve substrate stability.

Research Applications

This quantitative immunoassay is intended for research use only in human serum, supporting studies of fatty liver disease, metabolic syndrome, obesity-related hepatic complications, and pediatric NAFLD cohorts. Researchers investigating the genetic basis of liver disease benefit from the kit's standardized calibrator and ready-to-use reagent format. For research use only; not intended for diagnostic procedures.
