Human PMEL ELISA Kit

A sandwich ELISA from Assay Genie for the quantitative detection of human PMEL in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

PMEL (also known as PMEL17, P100, P1, SILV, ME20-M and Melanocyte protein Pmel 17) is a premelanosome protein involved in melanosome biogenesis and melanin synthesis, and its dysregulation has been associated with skin disorders and melanoma, supporting its use as a biomarker in metabolism research.

The workflow uses a biotin–streptavidin–HRP detection system: equilibrate reagents and TMB substrate, incubate standards and samples at 37°C for 90 minutes, add biotin-labelled detection antibody for 60 minutes, incubate with HRP-Streptavidin (SABC) for 30 minutes, develop with TMB in the dark for 10–20 minutes, and read absorbance at 450 nm after stop solution. Linearity across 1:2, 1:4 and 1:8 dilutions was 91–103%, 87–105% and 86–101% in serum; 85–94%, 93–101% and 86–100% in EDTA plasma; and 81–100%, 85–99% and 83–99% in heparin plasma. Spike-and-recovery averaged 93% in serum, 96% in EDTA plasma and 93% in heparin plasma.
