Human PLK1/Serine/threonine- kinase PLK1 ELISA Kit

This sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of human PLK1 (Polo-like kinase 1), a serine/threonine kinase that acts as a key regulator of cell cycle progression and is involved in mitosis and the DNA damage response. Dysregulation of PLK1 has been linked to cancer development and progression.

The assay uses a double-antibody sandwich format with colorimetric detection and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is reported at 4.688 pg/mL with a measurable range of 7.813–500 pg/mL. Linearity (n = 5) for serum is 82–100%, 87–103% and 90–98% at 1:2, 1:4 and 1:8 dilutions; for EDTA plasma 82–98%, 82–98% and 85–105%; and for heparin plasma 82–101%, 87–101% and 82–99%. Spike-and-recovery averages are 94% in serum (range 87–101%), 96% in EDTA plasma (range 87–100%) and 97% in heparin plasma (range 94–99%).

The protocol includes primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-Streptavidin (SABC) incubation for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition. Kits are stored at 2–8°C for 12 months.
