Human PLA2G7/PAF-AH/Lp-PLA2 ELISA Kit

A sandwich ELISA kit from Assay Genie for the quantitative measurement of human PLA2G7 (also known as PAF-AH or Lp-PLA2) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The double-antibody format uses colorimetric detection with a TMB substrate, and absorbance is read at 450 nm. Sensitivity is 0.938 ng/mL and the standard range is 1.563–100 ng/mL.

Reported linearity for serum (n=5) is 85–104% at 1:2, 83–100% at 1:4, and 86–98% at 1:8; for EDTA plasma (n=5) 87–101%, 85–100%, 82–101%; and for heparin plasma (n=5) 87–96%, 85–94%, 83–94%. Mean spike-and-recovery is 96% from serum, 97% from EDTA plasma, and 94% from heparin plasma.

The protocol comprises reagent and TMB equilibration to room temperature, a 90-minute primary incubation at 37°C, biotinylated detection antibody binding for 60 minutes at 37°C, HRP-streptavidin binding for 30 minutes at 37°C, TMB color development in the dark for 10–20 minutes, and immediate absorbance measurement at 450 nm after stop solution addition. Kit components are stored at 2–8°C for 12 months.
