Human PKC delta/Protein Kinase C Delta ELISA Kit

From Assay Genie, this sandwich ELISA uses a double-antibody format for the accurate detection of Protein Kinase C Delta (PKC Delta) in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay employs colorimetric detection, and PKC Delta is described as a critical protein kinase involved in cellular processes including cell growth, proliferation, and survival, with dysregulation linked to cancer, cardiovascular diseases, and neurodegenerative disorders.

Reaction absorbance is read at 450 nm after TMB substrate development, and storage is at 2-8°C for 12 months. The procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples, blanks and a zero control onto the pre-coated plate, incubating at 37°C for 90 minutes for antigen binding, adding a biotin-labeled detection antibody with a 37°C incubation of 60 minutes, binding HRP-Streptavidin (SABC) for 30 minutes at 37°C, developing colour with TMB substrate in the dark for 10-20 minutes, then adding stop solution and reading absorbance at 450 nm immediately.

Linearity has been demonstrated across 1:2, 1:4 and 1:8 dilutions (n=5) for serum (87-98%, 83-101%, 82-99%), EDTA plasma (92-99%, 90-98%, 82-95%) and heparin plasma (85-96%, 82-97%, 86-100%). Reported recovery ranges are 93-105% (average 98%) for serum, 88-103% (average 96%) for EDTA plasma, and 85-103% (average 94%) for heparin plasma (n=5).
