PTTG1IP, also known as PTTG1-binding factor (PBF), is a protein originally characterized through its interaction with securin (PTTG1), and this colorimetric ELISA Kit from Assay Genie provides quantitative measurement of the human form in tissue homogenates and other research specimens. Altered PTTG1IP expression has been investigated in pituitary tumors, thyroid carcinoma, and breast cancer, where it has been linked to cell proliferation, invasive cell behavior, and the regulation of iodide transport.

The assay is read on a standard microplate reader using a colorimetric endpoint, and each kit supplies reagents sufficient for 96 assays. Included components typically comprise a pre-coated microplate, matched capture and detection antibodies, a recombinant calibrator, wash buffer, substrate, and stop solution. Standard curves are generated from serial dilutions of the supplied reference standard, with unknown sample concentrations interpolated from absorbance values.

For sample preparation, fresh or frozen tissue is disrupted in an appropriate lysis buffer and clarified before loading. Reconstituted standards should be aliquoted and stored frozen to limit freeze-thaw loss, while working buffers are kept refrigerated until use. Because the antibodies were raised against the human sequence, cross-reactivity with non-human species has not been formally established and should be evaluated by the investigator before use in other models.
