ZAP70 (zeta-chain-associated protein kinase 70) is a Syk-family cytoplasmic tyrosine kinase that is essential for T-cell receptor (TCR) signal transduction. Following TCR engagement, ZAP70 is recruited to the immune synapse and activated through phosphorylation of regulatory tyrosine residues, including Y315, Y319, and Y493, which then propagate downstream signaling through adaptor proteins such as LAT and SLP-76.

This phosphorylation ELISA kit from Assay Genie is configured to measure phosphotyrosine content on ZAP70 in cell lysates and tissue lysates, supporting studies of T-cell activation, thymic selection, and adaptive immune signaling. ZAP70 is clinically relevant: autosomal recessive ZAP70 deficiency causes a form of severe combined immunodeficiency (SCID), while dysregulated ZAP70 activity has been implicated in T-cell acute lymphoblastic leukemia, chronic lymphocytic leukemia, and several autoimmune disorders.

Typical applications include characterization of TCR pathway activation in primary lymphocytes and Jurkat or other T-cell line models, time-course studies of immune cell signaling, and screening of small-molecule kinase inhibitors in immunology and immuno-oncology research. Each kit is supplied in a 96 assays format and is intended for research use only.
