The Human Phosphotyrosine ITK PharmaGenie ELISA Kit from Assay Genie is built around the interleukin-2-inducible T-cell kinase (ITK), a Tec-family non-receptor tyrosine kinase that becomes phosphorylated following T-cell receptor engagement. The microplate is pre-coated with an anti-ITK capture antibody so researchers can pull the kinase out of prepared cell lysates or tissue lysates and then probe the captured protein with a phosphotyrosine-specific detection antibody. This setup makes the kit useful for tracking activation-state changes downstream of TCR, CD28, and chemokine-receptor stimulation in primary human T cells, Jurkat lines, or differentiated Th2/Th17 cultures.

Typical workflows begin with stimulating cells, lysing them in a buffer containing protease and phosphatase inhibitors, and loading 100 uL of clarified lysate per well. Researchers running dose-response or time-course experiments frequently aliquot lysates and freeze them at -80 degrees C to limit freeze-thaw cycles, because phosphotyrosine levels decay rapidly once phosphatases are no longer inhibited. The phospho-specific ELISA readout complements total-ITK western blots when the goal is to quantify activation kinetics, screen small-molecule kinase inhibitors, or compare signaling across patient-derived T-cell samples in immunology, immuno-oncology, and transplant-tolerance research programs.
