The Assay Genie phosphofructokinase muscle isozyme (PFKM) immunoassay is a quantitative enzyme-linked immunosorbent assay built for measuring PFKM protein levels in human tissue homogenates and related samples. PFKM is the muscle-expressed isoform of phosphofructokinase-1, the rate-limiting glycolytic enzyme that phosphorylates fructose-6-phosphate to fructose-1,6-bisphosphate. Because PFKM dominates skeletal muscle and cardiac tissue, the assay supports studies of muscle glycogen metabolism, exercise physiology, and the pathophysiology of Tarui disease (glycogen storage disease type VII), in which inherited PFKM deficiency causes hemolytic anemia and myopathy. Researchers also deploy the kit to track PFKM expression in metabolic, cancer, and mitochondrial disorder models where glycolytic flux is of interest. Each kit supplies reagents and buffers sufficient for one microplate run of 96 determinations in a strip-well plate configuration, read on standard ELISA plate readers. Capture and detection antibodies are pre-titrated, reducing assay-development time. The assay follows a standard incubation and wash protocol requiring refrigerated storage of kit components and small microliter pipetting volumes, making it compatible with limited tissue biopsies and cell lysates.
