The Phospho-ZAP70 (Y493) and Total ZAP70 dual-analyte immunoassay from Assay Genie enables simultaneous quantification of activated ZAP70 kinase and total ZAP70 protein across the supplied microplate. ZAP70 is a 70 kDa Syk-family tyrosine kinase that couples T-cell receptor engagement to downstream signaling cascades; phosphorylation at the activation-loop residue Y493 is a direct readout of TCR-induced kinase activation. By measuring phospho-Y493 alongside total ZAP70 in parallel wells, researchers can normalize signaling output to protein expression, controlling for variations in T-cell numbers, transfection efficiency, or treatment-induced changes in total kinase levels. The kit accepts cell lysates and tissue lysates prepared with standard RIPA or NP-40 buffers supplemented with phosphatase inhibitors, and the strip plates are pre-coated with capture antibodies specific for human ZAP70, providing a human-specific measurement. Components should be refrigerated upon arrival and protected from light to preserve antibody binding activity.
