WNK1 (With-No-Lysine Kinase 1) is a serine/threonine kinase whose autophosphorylation at threonine 60 drives activation of the SPAK and OSR1 kinases, which in turn phosphorylate the NCC cotransporter that governs renal sodium reabsorption. Researchers studying hypertension, nephron ion transport, or kinase pathway regulation load cell lysates and tissue lysates to compare phosphorylated kinase against total expression across treatments, often quantifying percent phosphorylation relative to total protein. The colorimetric readout uses a standard microplate reader, and the 96 assays kit format provides sufficient wells to run both the phospho-specific and total target plates alongside a standard curve on a single plate.
