The Human Phospho-VEGFR2 (Y996) PharmaGenie ELISA Kit from Assay Genie is a phosphorylation-site-specific immunoassay that quantitatively measures tyrosine 996 phosphorylation of VEGFR2 (KDR/Flk-1), the major VEGF-A-driven autophosphorylation site that activates downstream PLCγ, MAPK, and PI3K/Akt signaling in vascular endothelial cells.

Because Y996 phosphorylation reports on receptor activation status rather than total receptor abundance, the kit is commonly used to profile angiogenic signaling in tumor microenvironment studies, retinal and choroidal neovascularization models, vascular permeability assays, and endothelial cell responses to candidate anti-angiogenic compounds.

  • Reactivity: human VEGFR2 phosphorylated at the Y996 epitope
  • Compatible samples: cell lysates and tissue lysates prepared in RIPA buffer supplemented with protease and phosphatase inhibitor cocktails to preserve phosphorylation
  • Assay format: 96-well microplate strip plate with pre-coated capture antibody
  • Detection: colorimetric readout on a standard plate reader at 450 nm
  • Sample volume: 100 µL of clarified lysate per well
  • Storage: store kit components at -20°C; thaw only the working aliquot and avoid repeated freeze-thaw of the phospho-specific detection antibody

The assay is intended for research use only and is best paired with a total VEGFR2 ELISA so investigators can calculate phospho/total ratios, monitor time-course activation by VEGF stimulation, and benchmark inhibitor potency across compound series.
