Developed by Assay Genie for quantitative measurement of TAK1 activation in human samples, this dual-analyte immunoassay simultaneously quantifies TAK1 phosphorylated at Ser412 alongside total TAK1 protein. Researchers working on NF-κB and MAPK signaling cascades can directly normalize phospho-TAK1 levels against total TAK1 from the same lysate, eliminating inter-well variability caused by differing protein loads. Ser412 is a key activating autophosphorylation site on TAK1 (MAP3K7), the upstream serine/threonine kinase that transduces signals from TNF-α, IL-1β, and Toll-like receptors. The kit accommodates cell lysates and tissue lysates, making it applicable to studies of inflammation, innate immunity, cellular stress, and cancer pathways. Absorbance is read on a standard microplate reader. Reagents are supplied on cold packs and components requiring long-term storage should be kept at -20 °C with limited freeze-thaw cycles to preserve antibody activity. The plate provides parallel phospho-specific and pan-TAK1 capture wells, supporting both endpoint and time-course signaling experiments across independent biological replicates.
