Human Phospho-STAT6 (Y641) and Total STAT6 PharmaGenie ELISA Kit (SBRS1995)

This dual-analyte ELISA kit from Assay Genie enables parallel measurement of STAT6 phosphorylated at tyrosine 641 and total STAT6 protein in the same lysate, supporting direct normalization of pathway activation against total STAT6 expression. Phosphorylation at Y641 is the activating event downstream of IL-4 and IL-13 receptor engagement, so this assay is widely applied in studies of Th2 lymphocyte differentiation, IgE class-switch recombination, allergic airway inflammation, asthma pathogenesis, and STAT6-dependent oncology research including Hodgkin lymphoma and colorectal cancer signaling.

The kit is configured for 96 reactions per plate in microplate format and is validated for cell lysate and tissue lysate inputs, allowing direct assessment of signaling status from cultured cells or homogenized tissue without requiring serum or plasma samples. Each well receives 100 uL of prepared lysate, and lysates should be prepared with protease and phosphatase inhibitor cocktails to preserve the Y641 phospho-epitope.

Included reagents are supplied in concentrated form along with assay buffer, antibody-coated plate, detection antibody, and substrate solution. Store all components refrigerated between runs, protect substrate from light, and minimize freeze-thaw cycles of the phospho-specific detection antibody to preserve binding activity.
