This dual-analyte ELISA from Assay Genie quantifies SRC tyrosine kinase activation by simultaneously measuring phosphorylation at the activating residue Y419 within the kinase activation loop alongside total SRC protein levels. Capturing phospho-SRC (Y419) and total SRC in matched wells enables direct normalization of phosphorylation status to total kinase expression, supporting accurate comparison of SRC activation across treated versus untreated samples.

SRC is a non-receptor tyrosine kinase central to signaling pathways that control cellular proliferation, migration, adhesion, and survival. Aberrant SRC activity at Y419 has been implicated in tumor progression, invasion, and kinase inhibitor resistance, making quantitative phosphorylation measurements valuable in oncology, signal transduction, and drug-discovery research. The antibodies are specific to human SRC and show minimal cross-reactivity with rodent orthologs, and the kit accepts cell lysates and tissue lysates prepared with standard lysis buffers containing phosphatase inhibitors to preserve the Y419 phospho-epitope.

Each kit provides a pre-coated microplate, capture and detection antibodies, recombinant phospho-SRC and total SRC protein standards, and all reagents required for absorbance-based readout, allowing processing of up to 96 assays per plate. Reagents should be refrigerated upon arrival and stored according to the supplied stability data. Results are reported as a ratio of phospho-SRC (Y419) to total SRC, reflecting true kinase activation state for research use only.
