SMAD5 is a receptor-regulated SMAD protein that transduces bone morphogenetic protein (BMP) signals from the cell surface to the nucleus. Phosphorylation at Ser463/465 is mediated by BMP type I receptors (ALK2, ALK3, and ALK6) and triggers SMAD5 complex formation with SMAD4, driving transcription of BMP-responsive genes involved in osteogenesis, iron homeostasis through hepcidin regulation, and vascular remodeling.

This dual-capture ELISA from Assay Genie simultaneously quantifies phosphorylated SMAD5 at Ser463/465 and total SMAD5 levels in human cell lysates and tissue lysates, enabling researchers to normalize pathway activation against total protein abundance. The assay produces a colorimetric readout and is supplied as a 96-assay kit suitable for signaling studies, compound screening, and characterization of BMP pathway modulators in oncology, hematopoiesis, and skeletal biology research.

Each kit contains pre-coated plates, site-specific and pan-SMAD5 detection antibodies, recombinant protein standards, dilution buffers, wash buffer, and substrate reagents. Researchers commonly pair this ELISA with Western blot validation of SMAD5 phosphorylation in BMP2- or BMP4-stimulated cells, in studies of bone formation and fracture repair, in models of hereditary hemorrhagic telangiectasia, and in erythropoiesis research where SMAD5 is required for normal red blood cell development.
