Human Phospho-SMAD4 (Thr277) PharmaGenie ELISA Kit (SBRS1972)

The Human Phospho-SMAD4 (Thr277) PharmaGenie ELISA Kit from Assay Genie is a phosphorylation-specific immunoassay engineered to quantify SMAD4 protein phosphorylated at threonine 277. The capture and detection antibodies are specific for the human SMAD4 ortholog and have been validated against phosphorylated Thr277, making the kit applicable to studies of canonical SMAD pathway activity in cultured mammalian cells and in tumor or biopsy-derived material. The assay accepts cell lysates and tissue lysates as input.

SMAD4 (also known as DPC4) functions as the common-mediator SMAD that partners with receptor-activated SMADs (SMAD2 and SMAD3 for TGF-beta pathways; SMAD1, SMAD5 and SMAD8 for BMP pathways) to translocate into the nucleus and regulate target-gene transcription. Phosphorylation at Thr277 has been characterized in downstream signaling events and is of particular interest in pancreatic, colorectal and gastrointestinal cancer research, where SMAD4 loss-of-function or pathway rewiring is a frequent feature.

Researchers use this kit to monitor pathway activation in response to ligand stimulation (TGF-beta1, BMP2, activin), small-molecule inhibitors (kinase inhibitors, receptor tyrosine kinase blockers), or genetic manipulation (siRNA, CRISPR knockout of upstream kinases). The kit is supplied as a complete set of reagents sufficient for 96 individual determinations, including a pre-coated microplate, calibration standards, detection antibody and assay buffer. Store unused components under refrigerated conditions (do not freeze the pre-coated plate); for longer-term storage of standard and antibody vials, aliquot and keep at -20 degrees C, limiting freeze-thaw cycles. Reconstituted working solutions should be consumed within the stability window printed on the lot-specific certificate of analysis.
