Phospho-SMAD1 (Ser463/465) sits at the heart of canonical BMP (Bone Morphogenetic Protein) signalling, where activated type I BMP receptors phosphorylate SMAD1 at this C-terminal motif to drive nuclear translocation and target-gene transcription. Quantifying this modification with Assay Genie's phosphorylation ELISA lets researchers track pathway activation in response to BMP2, BMP4, BMP6, BMP7 or small-molecule agonists and inhibitors in osteoblast, chondrocyte, neural stem cell and hepatic hepcidin-regulation studies.
Validated for human cell lysates and tissue lysates, the kit is supplied as a microplate with reagents for 96 assays and a recommended lysate sample volume of 100 µL per well. Store the reconstituted standards and detection antibody frozen at -20°C in working aliquots to limit freeze-thaw cycles; other components should be kept refrigerated and protected from light. Intended for research use only, this ELISA complements western blot validation, BMP-driven osteogenic differentiation studies, and signalling cross-talk investigations across the TGF-β superfamily.
