This phosphorylation ELISA from Assay Genie simultaneously quantifies SMAD1 phosphorylated at Ser463/465 alongside total SMAD1 protein in a single well, enabling researchers to normalize BMP pathway activation to total protein expression. SMAD1 is a downstream effector of BMP type I receptors, and phosphorylation at these serine residues is a direct indicator of canonical bone morphogenetic protein signaling activity. The kit is validated for use with cell lysates and tissue lysates from human, mouse, and rat samples, supporting applications such as studies of osteoblast differentiation, stem cell lineage commitment, and TGF-β superfamily crosstalk. Detection uses a colorimetric readout on a microplate, with results expressed as a ratio of phosphorylated to total SMAD1 to control for variation in protein loading across samples. Each well requires 100 µL of sample, and the kit provides sufficient reagents for 96 individual reactions. Reagents should be stored refrigerated at 2-8°C.
