Description

The SHIP1 phosphatase (encoded by INPP5D) is a negative regulator of phosphoinositide signaling, dephosphorylating PI(3,4,5)P3 at the 5-position to dampen PI3K-driven AKT activation in hematopoietic cells. Tyrosine 1020 lies in the C-terminal proline-rich domain, and phosphorylation at this site marks receptor-proximal activation downstream of BCR, FcR, and cytokine receptor engagement. This Assay Genie dual-analyte ELISA resolves phospho-SHIP1 (Y1020) against total SHIP1 on a single microplate, allowing normalization across treatments without parallel Western blot runs.

Validated for in vitro research use only on cell lysates and tissue lysates, the kit relies on human-specific capture antibodies and shows no significant cross-reactivity with rodent SHIP1 orthologs. A typical input of 5 to 10 ug of total protein per well is sufficient, and reconstituted standards should be aliquoted and stored frozen to limit freeze-thaw cycles; unopened components are kept refrigerated. The kit includes a strip plate, lyophilized standards, detection antibodies, wash buffer, and substrate. Researchers studying B-cell receptor signaling, lymphoma biology, or myeloid checkpoint pathways can pair this readout with upstream kinase assays and downstream phospho-AKT measurements.
