Phospho-PRAS40 (T246) and total PRAS40 are quantified from the same sample lysate, enabling researchers to normalize phosphorylation status to total protein expression. PRAS40 (Proline-Rich AKT Substrate of 40 kDa), also known as AKT1S1, is phosphorylated at threonine 246 by activated AKT, and this modification relieves its inhibitory binding to mTORC1, making the phospho-PRAS40/total-PRAS40 ratio a downstream readout of PI3K-AKT pathway activity. The Assay Genie dual-analyte format is therefore useful in studies of insulin signaling, oncogenic AKT activation, and the cellular response to kinase inhibitors or growth factor stimulation, with antibodies validated on human and mouse cell lysates.

Each kit provides pre-coated microwell plates along with calibrated standards and detection antibodies for both the phosphorylated and total targets.
