Quantitative measurement of threonine-495 phosphorylation on PKMYT1 (also called MYT1) in human samples is enabled by this immunoassay from Assay Genie. PKMYT1 is a membrane-associated serine/threonine kinase that restrains CDK1 activity during the G2/M checkpoint by phosphorylating Thr14 and Tyr15 on CDK1, and the T495 site on PKMYT1 itself is targeted by upstream regulators such as casein kinase 2 and CDK1-cyclin B. Studying this modification is relevant to DNA damage response research, mitotic entry control, and investigations of Wee1-family inhibitor efficacy in cancer models. The kit is configured for use with cell lysates and tissue lysates prepared by standard RIPA or NP-40-based protocols, and is supplied with pre-coated plates, detection antibody, buffers, and a recombinant phospho-protein standard required to build the standard curve. Each pack contains reagents for 96 individual assays and is intended for research use only in laboratory settings.
