The Phospho-PAK2 (Ser20) and Total PAK2 ELISA Kit from Assay Genie enables simultaneous quantification of the phosphorylated Ser20 form and total p21-activated kinase 2 protein within a single microplate workflow. PAK2 is a serine/threonine kinase regulated by the Rho-family GTPases Rac1 and Cdc42, and the Ser20 residue sits within the N-terminal regulatory domain where phosphorylation couples to cytoskeletal reorganization, membrane ruffling, focal adhesion turnover, and caspase-3-mediated apoptotic signaling. Researchers studying motility, growth factor responses, oxidative stress, or chemotherapeutic-induced apoptosis in human cellular models can use this dual readout to monitor activation-state changes that would be invisible in total-protein-only assays.
For sample preparation, lyse cultured cells or freshly harvested tissue in a buffer containing protease and phosphatase inhibitors to preserve the Ser20 phosphorylation mark, then clarify by centrifugation at 12000 x g for 10 minutes. Load approximately 100 uL of diluted lysate per well alongside the supplied phospho- and total-protein reference standards, allowing direct normalization of phospho-PAK2 signal to total PAK2 abundance in each well. The assay is intended for research use only in vitro applications and is not validated for diagnostic or clinical specimen testing.
Upon arrival, store unopened components refrigerated and keep the conjugated detection reagent protected from light. Reconstituted standards and any unused diluted samples should be aliquoted and frozen at -80 °C to limit freeze-thaw cycles, since repeatedly thawed reagents show reduced signal. The microplate strip format allows running partial plates when sample numbers are limited, and the kit supports standard curve concentrations spanning the low ng/mL range, giving sufficient dynamic range to detect modest pathway activation changes in treated versus control lysates.
