The Phospho-P27/Kip1 (T198) ELISA Kit from Assay Genie enables quantitative measurement of the threonine-198 phosphorylated form of the cyclin-dependent kinase inhibitor p27 (CDKN1B) in cell and tissue lysates. T198 phosphorylation is a downstream readout of PI3K/Akt signaling and drives cytoplasmic relocalization of p27, where it acquires pro-oncogenic functions tied to cytoskeletal remodeling, cell migration, and invasion. Investigators in oncology, particularly breast, prostate, and colorectal cancer research, use T198-phospho-p27 quantification to monitor pathway engagement and characterize responses to kinase inhibitors and targeted therapeutics.

The pre-coated well plate provides enough capacity for individual sample determinations and is read on a standard microplate reader. Each well is pre-coated with a capture antibody recognizing total human p27, while the detection reagent specifically recognizes the T198-phosphorylated epitope. A recombinant phospho-peptide standard curve is supplied for interpolating absorbance values. Reconstituted standards and detection antibody should be stored at -20°C in single-use aliquots to limit freeze-thaw cycles; the coated plate is kept refrigerated at 4°C until first use.

Validated for human samples; cross-reactivity with rodent p27 orthologs has not been characterized in the product insert, so xenograft and murine tissue lysates should be confirmed by orthogonal methods such as western blot or immunohistochemistry. Intended for research use only, not for diagnostic procedures.
