Dual-Target Phosphoprotein Profiling

This Assay Genie kit delivers simultaneous quantification of the activated and baseline pools of MSK2 in one run. The microplate is partitioned so that wells reserved for phospho-Ser360 signal and wells reserved for total MSK2 are developed side by side, allowing end-users to derive a normalized pMSK2-to-total ratio and correct for well-to-well loading differences that often confound single-analyte measurements.

Biological Role of MSK2 S360 Phosphorylation

MSK2 (RPS6KA4) is a nuclear serine/threonine kinase downstream of the ERK1/2 and p38 MAPK cascades. Serine-360 sits within its activation loop; modification here has been linked in peer-reviewed work to chromatin remodeling, immediate-early gene induction, and stress responses across models of inflammation, corticosteroid signaling, and oncogenic rewiring. Detecting this mark quantitatively is therefore central to studies of mitogenic and genotoxic signaling networks.

Workflow and Sample Requirements

Each well is loaded with a 100 uL clarified lysate prepared from cultured cells or snap-frozen tissue. Capture antibodies are designed for human samples and show limited cross-reactivity with rodent orthologues, which makes the workflow well-matched to patient-derived material, tumor lines, and primary human cultures. Following incubation with HRP-conjugated detection reagent and chromogenic substrate, signals are read against a parallel standard curve of recombinant phospho- and total-MSK2 protein, enabling accurate back-calculation of the phospho-Ser360 fraction.
