The Human Phospho-MSK2 (S360) PharmaGenie ELISA Kit from Assay Genie is a phosphorylation-specific immunoassay for quantitative detection of mitogen- and stress-activated protein kinase 2 (MSK2, also known as RSK-B / RPS6KA4) phosphorylated at serine 360. MSK2 is a nuclear AGC-family kinase activated downstream of the ERK1/2 and p38 MAPK cascades in response to growth factors, oxidative stress, UV, and inflammatory cytokines, and S360 phosphorylation is a key activation marker used in studies of chromatin remodeling, CREB and ATF1 transcription factor regulation, and immediate-early gene expression.

The kit is supplied on a pre-coated microplate and uses a phospho-S360-selective detection antibody to discriminate the activated pool of MSK2 from total kinase in human cell lysates and tissue lysates. Common applications include time-course and dose-response experiments in cancer cell lines, primary neurons, and stimulated immune cells where researchers track pathway activation following cytokine or stress treatments. Each well uses 100 µL of prepared lysate as the sample volume, and the included standard curve enables quantitative comparison of phospho-MSK2 levels across treatment conditions.

For stability, unopened components should be stored refrigerated; working aliquots of standards and detection antibody are best kept frozen at -20°C and protected from light, with repeated freeze-thaw cycles avoided to preserve antibody binding activity. The kit format includes pre-coated wells, detection antibody, dilution and wash buffers, TMB substrate, and stop solution in a research-use-only configuration.
