This Phospho-MSK1 (S376) and Total MSK1 ELISA from Assay Genie enables quantification of endogenous MSK1 kinase activity in cell and tissue lysates. MSK1, also known as RPS6KA5, operates downstream of the ERK1/2 and p38 MAPK pathways; phosphorylation at serine 376 within its C-terminal autoinhibitory domain marks the fully activated form of the kinase.

Reagents are supplied as a complete microplate with matched antibody pairs that resolve the phospho-S376 epitope from total MSK1 protein on the same lysate, enabling normalization of pathway activation to total kinase expression. The capture antibodies are raised against the human RPS6KA5 sequence, and the assay is reactive with human samples. Recommended applications include monitoring growth factor, cytokine, or stress-induced MAPK signalling, evaluating downstream CREB and histone H3 Ser10 phosphorylation, and screening kinase inhibitor compounds in vitro.
