LAT (Linker for Activation of T cells) is a 36-38 kDa transmembrane adaptor protein that scaffolds signaling intermediates downstream of the T-cell receptor. Tyrosine 161 is a key ZAP-70 substrate site whose phosphorylation drives calcium mobilization, MAPK activation, and immune synapse formation. This dual phospho/total format from Assay Genie lets researchers normalize site-specific phosphorylation against total protein abundance, a critical control in signaling studies where expression varies between cell states and treatment conditions.

The kit accommodates 96 assays using cell lysates or tissue lysates prepared with protease and phosphatase inhibitor cocktails to preserve the Y161 mark, which is otherwise rapidly removed by cellular phosphatases. Validated on human peripheral blood mononuclear cells and mouse splenocytes, the assay delivers quantitative readouts with sensitivity calibrated below 0.1 ng/mL for phospho-LAT (Y161). Investigators studying T-cell activation, anergy, immune checkpoint blockade, and CAR-T cell function use this assay to quantify signaling competence at the LAT node. Aliquot reconstituted reagents and freeze to maintain phospho-epitope activity across repeated experiments.
