The Phospho-JAK2 (Y1007/1008) ELISA kit from Assay Genie quantitatively measures JAK2 phosphorylated at the activating tyrosine residues 1007 and 1008 within the kinase activation loop. These two adjacent tyrosines are trans-autophosphorylated upon cytokine receptor engagement, and their modification is the primary biochemical read-out of JAK2 catalytic activation. The kit is intended for research use only and supports studies of the JAK-STAT signaling cascade, including work on cytokine-driven hematopoiesis and myeloproliferative neoplasms such as polycythemia vera and essential thrombocythemia, where the V617F gain-of-function mutation drives constitutive phosphorylation at these sites.

Untreated and stimulated cell or tissue lysates can be compared directly, supporting dose-response and inhibitor studies with small-molecule JAK inhibitors such as ruxolitinib and fedratinib. Samples are prepared in lysis buffer compatible with phosphoprotein preservation, and the well plate is processed at room temperature using a standard ELISA workflow with washing steps between incubations. Capture of phosphorylated JAK2 relies on a site-specific antibody recognizing the dual phospho-epitope, allowing discrimination between catalytically active kinase and total JAK2 protein levels. Reagents should be kept refrigerated; frozen aliquots of standards and antibodies are best held at -20 °C or below to preserve phospho-epitope integrity, with limited freeze-thaw cycles recommended.
