Phospho-IκB alpha (Ser32) sits at a critical control point in the canonical NF-κB signaling cascade. When the IKK complex is activated by upstream stimuli such as TNF-α, IL-1β, or LPS, it phosphorylates IκBα on Ser32, triggering ubiquitination and proteasomal degradation so that NF-κB dimers can translocate to the nucleus and drive inflammatory gene transcription. The Assay Genie Human Phospho-I-kappa B alpha (S32) PharmaGenie ELISA Kit is built to quantify that phosphorylation event directly in human cell lysates and tissue lysates, supporting pathway studies that monitor activation kinetics, compound screening against IKK or upstream kinases, and comparison of basal versus stimulated states.

The kit ships as a 96-assay microplate in the PharmaGenie reagent line, processed with standard microplate workflows and read on a conventional plate reader using a colorimetric endpoint. Because the capture reagent recognizes the phosphorylated epitope rather than total IκBα, the readout distinguishes signaling-driven modification from steady-state protein levels, an important control when interpreting pathway activation. Cell lysate and tissue lysate inputs accommodate adherent and suspension culture work as well as snap-frozen tissue biopsies from inflammation, oncology, and immunology models.

Typical research applications include monitoring NF-κB pathway engagement downstream of cytokine or pathogen-associated molecular pattern challenge, evaluating candidate IKK inhibitors, profiling tumor samples for constitutive pathway activity, and time-course studies of signaling decay following pharmacologic intervention. The 96-assay configuration provides enough wells for multi-condition designs, including dose-response curves, kinetic series, and matched stimulated-versus-rested sample pairs.
