The Human Phospho-Glucocorticoid Receptor (S226) and Total Glucocorticoid Receptor ELISA from Assay Genie enables simultaneous quantification of phosphorylated GR at serine 226 alongside total GR protein within the same sample. This dual-analyte format allows researchers to normalize phosphorylation levels to total receptor expression, providing a more accurate readout of glucocorticoid receptor activation state under varied experimental conditions.

Serine 226 phosphorylation is a well-characterized regulatory modification of the glucocorticoid receptor, influenced by MAPK-dependent signaling cascades and modulated by ligand binding, cellular stress, and inflammatory stimuli. The assay is validated for use with cell lysates and tissue lysates, with a recommended sample volume of 100 μL per well, and shows species-specific reactivity with human glucocorticoid receptor, making it applicable to studies of glucocorticoid signaling in hormone-responsive tissues, immune cell models, and research into stress responses, inflammation, and metabolic regulation.

Supplied as a complete microplate-based assay kit with all necessary reagents, the format supports 96 assays per run. Lysate samples should be handled according to standard lysis protocols and stored refrigerated at 4 degrees C for short-term use or frozen at -20 degrees C for longer preservation to maintain the integrity of the phospho-epitope.
