The Human Phospho-FYN (Y530) and Total FYN ELISA Kit from Assay Genie provides a dual-analyte immunoassay that simultaneously quantifies the level of FYN phosphorylated at tyrosine 530 alongside total FYN protein within a single sample. This phospho-to-total pairing allows researchers to normalize site-specific phosphorylation against total kinase expression, correcting for differences in protein loading or expression between experimental conditions.

FYN is a 59 kDa non-receptor tyrosine kinase of the Src family, broadly expressed in immune and neural tissues. Tyrosine 530 sits in the C-terminal tail and is phosphorylated by the CSK kinase; this modification stabilizes a closed, autoinhibited conformation. Dephosphorylation of Y530 releases the intramolecular SH2 and SH3 domain interactions, activating FYN's catalytic function. The kit therefore reports on the balance between CSK-mediated repression and upstream phosphatase activity in cell signaling studies.

Applications include investigation of T-cell receptor signaling, integrin and focal-adhesion pathways, cytoskeletal regulation, and FYN's role in neuronal development and oncology research. The assay accepts cell lysates and tissue lysates prepared with phosphatase inhibitors to preserve the Y530 modification. Capture antibodies are provided in a microplate format for parallel processing of multiple conditions, with a standard curve for quantitative interpolation. Reagents should be stored at -20 degrees C; allow the microplate and buffers to equilibrate to room temperature before running the assay.
