Human Phospho-FLT3 (Y589) PharmaGenie ELISA Kit (SBRS1829)

FLT3 (FMS-like tyrosine kinase 3) is a class III receptor tyrosine kinase essential for hematopoietic progenitor proliferation and differentiation. Tyrosine 589 sits within the activation loop of the kinase domain and acts as a key autophosphorylation site, reporting ligand-driven activation as well as signaling output from the internal tandem duplication (ITD) and tyrosine kinase domain (TKD) mutations frequently observed in acute myeloid leukemia. This phospho-specific ELISA from Assay Genie enables direct quantification of Y589 phosphorylation status independent of total receptor expression, supporting work on AML biology, kinase inhibitor pharmacology, and downstream STAT5, MAPK, and PI3K pathway engagement.

Each kit ships with a pre-coated microplate, anti-phospho-FLT3 capture antibody, detection reagents, and diluents sufficient for a full plate of cell lysate and tissue lysate samples, loaded with a 100 uL sample volume per well. Lysis buffer should be freshly supplemented with phosphatase and protease inhibitors, and working aliquots are best stored frozen, with limited freeze-thaw cycles to preserve the phospho-epitope. Standard curve wells, sample wells, and blank wells are processed in parallel on the same plate, allowing direct interpolation of phosphorylation intensity across treatment conditions.
