Assay Genie's Phospho-FLT3 (Y589) and Total FLT3 PharmaGenie ELISA Kit is built for researchers studying receptor tyrosine kinase signaling in hematopoietic malignancies. FLT3 (CD135) is a class III receptor tyrosine kinase essential to early hematopoietic progenitor survival and proliferation, and activating mutations — most notably internal tandem duplications (FLT3-ITD) — are among the most frequent genetic alterations in acute myeloid leukemia, driving constitutive kinase activation.

Tyrosine 589 sits within the FLT3 juxtamembrane domain and acts as a major autophosphorylation site that couples receptor engagement to downstream PI3K/AKT and RAS/MAPK cascades. Quantifying phosphorylation at this residue alongside total FLT3 protein allows normalization across samples, separating true receptor activation from changes in expression alone.

Validated for human cell lysates and tissue lysates with reactivity to human FLT3 (CD135), the dual-analyte format is suited to in vitro profiling of leukemia cell lines, primary AML blasts, and FLT3-transfected model systems. Common applications include comparing basal versus ligand-stimulated phosphorylation with recombinant FLT3 ligand, evaluating tyrosine kinase inhibitor potency (quizartinib, midostaurin, gilteritinib), and tracking signaling dynamics following CRISPR, RNAi, or small-molecule intervention.

Each microplate delivers absorbance-based detection. Reconstituted standards, capture antibody, detection antibody, and buffers should be refrigerated and handled per the supplied protocol to preserve binding activity across the included 96-assay runs. For research use only — not intended for diagnostic procedures.
