Phospho-FAK (Y397) refers to Focal Adhesion Kinase phosphorylated at tyrosine 397, the primary autophosphorylation site that activates FAK signaling and drives integrin-mediated cell adhesion, migration, and survival pathways.

Researchers commonly use this Assay Genie immunoassay to quantify phosphorylation-level changes in response to stimuli such as growth factors, ECM engagement, or pharmacological inhibitors in cancer, wound healing, and mechanotransduction studies.

The kit quantifies activated FAK directly in cell lysates and tissue lysates using a colorimetric microplate ELISA format. Detection relies on a phospho-specific capture antibody that recognizes the Y397 epitope exclusively when phosphorylated, paired with a matched detection antibody.

Samples are prepared in lysis buffer supplemented with protease and phosphatase inhibitors to preserve the Y397 modification, which is labile once cells are disrupted.

This assay is intended for research use only and is widely cited in studies of tumor invasion, focal adhesion dynamics, and signal transduction.
