Phosphorylation of estrogen receptor alpha at serine 118 (ERα S118) is an established activation mark driven by MAPK/ERK signaling and serves to dissect ligand-dependent and ligand-independent ER activity in breast cancer research. This Assay Genie cell-based phosphorylation ELISA targets the phospho-S118 epitope, allowing researchers to quantify receptor activation directly in cultured cell lysates and frozen tissue lysates without the need for radioactive ATP labeling.

The kit is supplied as a complete 96-assay package with pre-coated microplate strip wells, phospho-specific capture antibody, detection reagents, and TMB substrate, generating a colorimetric signal readable on a standard plate reader. Each well is loaded with 100 μL of prepared sample lysate, and the included reconstituted standard and quality-control lysate allow pathway activation to be benchmarked between treatment conditions, time-course experiments, and receptor mutants. The in vitro assay workflow is compatible with standard laboratory lysate preparation protocols and has been validated on MCF-7 human breast carcinoma cell lysates as a positive reference.

For best stability, store all components at -20 °C and protect the substrate from light; avoid repeated freeze-thaw cycles of the working antibody and lyophilized standards.
