The Phospho-eIF-4E (S209) and Total eIF-4E dual-analyte format from Assay Genie enables researchers to quantify both the activated (serine 209 phosphorylated) and total pools of eukaryotic initiation factor 4E within the same experiment, with cross-reactivity validated against human, mouse, and rat eIF-4E, supporting direct calculation of phosphorylation stoichiometry without inter-assay variability.

eIF-4E Ser209 phosphorylation is catalyzed by MNK1 and MNK2 kinases downstream of MAPK and mTOR signaling, and elevated p-eIF-4E levels are implicated in oncogenic translation control, tumor progression, and therapy resistance mechanisms across multiple cancer models. By capturing lysate-derived protein on pre-coated microwells, this 96-well plate format kit delivers detection sensitivity in the low pg/mL range and supports studies in cap-dependent translation regulation, signal transduction pathway analysis, and pharmacological response profiling using cell lysates or tissue lysates as input material.

For in vitro research use, the assay provides a quantitative read-out of pathway activation state, making it useful for kinase inhibitor screening, target validation, and biomarker discovery workflows where comparing phospho versus total protein within each sample is essential.
