Built around a phosphorylation-site-specific antibody, this ELISA quantifies the activated form of the cAMP response element-binding protein, CREB, when it carries phosphate at serine 133. CREB is a calcium- and cAMP-responsive transcription factor, and S133 phosphorylation is the canonical switch that lets it recruit CBP/p300 and drive gene expression downstream of PKA, MAPK, and CaMKIV signaling. Tracking phospho-CREB (S133) is a standard readout in studies of neuronal plasticity, learning and memory, addiction pathways, GPCR-cAMP coupling, hormone signaling, and cellular stress responses.

Produced by Assay Genie as part of the PharmaGenie reagent line, the kit is configured for 96 assays per plate and validated for use with human cell lysates and human tissue lysates. The colorimetric workflow fits standard microplate readers, and the phospho-specific capture lets researchers separate the activated CREB pool from basal CREB in the same lysate, supporting parallel total-versus-phosphorylated comparisons when paired with a pan-CREB reagent.

Typical research applications include pathway-targeted compound screening, time-course and dose-response studies of kinase activators or inhibitors, validation of CRISPR or RNAi perturbations against upstream CREB kinases, and profiling of CREB pathway activation across patient-derived samples. The kit is intended for research use only. To preserve antibody and standard performance, store reagents as printed on the supplied insert, protect the streptavidin-HRP and substrate components from light, avoid repeated freeze-thaw cycles of the standard and antibody vials, and equilibrate the plate and buffers to room temperature before each run.
