Built around a phosphorylation-specific capture of Bruton's tyrosine kinase, this Assay Genie kit quantifies the Y551-autophosphorylated pool of BTK, the Tec-family kinase whose modification at the activation loop drives downstream phospholipase C-gamma signaling and calcium flux through the B-cell receptor cascade. Reading only the phospho-Y551 form lets laboratories separate constitutive BTK output from pathway-engaged BTK in experimental and patient-derived material, supporting work on B-cell malignancy biology, chronic lymphocytic leukemia response profiling, and the cellular mechanism of action of covalent BTK inhibitors such as ibrutinib and acalabrutinib.

Prepared cell lysates and homogenized tissue lysates are the recommended inputs, fitting in vitro stimulation experiments, ex vivo primary lymphocyte studies, and tumor biopsy or xenograft material. The protocol runs as an absorbance-based microplate immunoassay configured for 96 assay determinations per kit, so a single plate can carry a standard curve, internal controls, and unknowns together for batch comparison. Working antibody and conjugate solutions should be aliquoted to preserve phospho-epitope recognition across repeated freeze-thaw cycles, and the reagents are supplied for research use only.
