Description

A sandwich ELISA from Assay Genie for the quantitative detection of human LCAT (Phosphatidylcholine-sterol acyltransferase), also known as lecithin-cholesterol acyltransferase or phospholipid-cholesterol acyltransferase. The assay is intended for measuring LCAT in serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids. LCAT is a central enzyme in the extracellular metabolism of plasma lipoproteins, synthesised mainly in the liver and secreted into plasma, where it converts cholesterol and phosphatidylcholines (lecithins) to cholesteryl esters and lysophosphatidylcholines on the surface of HDL and LDL particles. Dysregulation of LCAT is associated with cardiovascular disease, supporting its use as a biomarker in cardiovascular research.

Performance is characterised by a detection range of 0.156-10ng/mL and a sensitivity of 0.098ng/mL. Reproducibility is reported with an intra-assay CV of 4.6% and an inter-assay CV of 8.3%. Specificity is confirmed against natural and recombinant human Phosphatidylcholine-sterol acyltransferase.

Linearity was assessed at dilutions of 1:2, 1:4, 1:8 and 1:16 (N=5) in serum, EDTA plasma and heparin plasma. Recovery is reported at 99% (range 93-105%) for serum and 101% (range 95-107%) for plasma. The kit is for research use only.
