For assay setup, dispense 100 µL of standards, controls, and prediluted samples into the pre-coated wells in duplicate, then add the biotinylated detection antibody and incubate at 37 °C for approximately 90 minutes so the sandwich complex can form. After a thorough wash step to remove unbound material, streptavidin-HRP conjugate is added and incubated briefly before a second wash. TMB substrate is then dispensed, allowing color to develop in the dark at room temperature for 15-20 minutes; the reaction is terminated with stop solution, shifting the chromogen from blue to yellow, and absorbance is read at 450 nm with a 570 or 630 nm reference filter for background correction.

The included lyophilized recombinant PGC standard is reconstituted to establish a seven-point standard curve, typically spanning roughly 0.78 to 50 ng/mL, which is fitted with a four-parameter logistic regression to interpolate unknown sample concentrations. Reported intra-assay precision is generally below 10% CV and inter-assay precision below 12% CV, and the analytical sensitivity of comparable Pepsinogen C immunoassays reaches sub-ng/mL detection limits. Because circulating PGC is more stable than PGI in serum, the analyte tolerates standard handling and short-term 4 °C storage, but repeated freeze-thaw cycles should be avoided.

Beyond the atrophic gastritis and gastric cancer research context already described, PGC quantification supports investigations into duodenal ulcer biology, Zollinger-Ellison syndrome, and pediatric gastric function studies, and is frequently combined with gastrin-17 (G-17) and anti-Helicobacter pylori IgG serology in published biomarker panels to stratify gastric cancer risk in population screening cohorts. Assay Genie also provides matched PGI and H. pylori antigen ELISA kits that can be ordered alongside this PGC kit for multi-analyte gastric mucosal profiling.
