Human PFN2/Profilin-2 ELISA Kit

Assay Genie's Human PFN2/Profilin-2 ELISA Kit is a sandwich ELISA using a double-antibody format and a colorimetric detection method, designed to quantify profilin 2 in human samples. The kit is reactive with human samples and supports research in cell biology, where profilin 2 is recognised for its role in regulating actin dynamics and cytoskeleton organisation, as well as in processes such as cell migration, proliferation, and intracellular signalling.

According to the manufacturer, the assay has a sensitivity of 0.375 ng/mL and a working range of 0.625–40 ng/mL. It is intended for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and the kit components should be stored at 2–8°C for 12 months. Reported linearity for serum (n=5) is 88–101% at 1:2, 85–101% at 1:4, and 93–105% at 1:8; for EDTA plasma (n=5), 82–97%, 88–97%, and 88–100% at the same dilutions; and for heparin plasma (n=5), 85–94%, 82–98%, and 80–98%. Recovery ranges are 88–101% (average 96%) for serum, 90–103% (average 98%) for EDTA plasma, and 86–102% (average 95%) for heparin plasma.

The assay procedure involves equilibrating reagents and TMB substrate, loading standards and samples for a 90-minute primary incubation at 37°C, binding with a biotin-labelled detection antibody for 60 minutes, incubating with HRP-Streptavidin (SABC) for 30 minutes, developing colour with TMB substrate for 10–20 minutes in the dark, and reading absorbance at 450 nm after adding stop solution.
